The Afonin lab has been provided with a seven-room laboratory (computational, wet-lab, bacterial lab, cold room, BSL-2 and BSL-2 plus cell culture rooms, and instrumentation room) on the second floor of the UNC Charlotte Science building. Lab space totals approximately 1550 sq ft and includes six sinks, a fume hood, shelf space, and exceptional bench and drawer space. The computational room totals approximately 350 sq feet and has 15 PCs and office space for students.
This laboratory is well-equipped to perform the synthesis and detailed characterization of nucleic-acid nanoparticles and lipoplexes. This laboratory contains a walk-in cold room with shelves and two Fisher Scientific FB300 power systems, two BioRad T100 Thermal Cyclers, a BioRad ChemiDoc MP Imaging system, a NanoDrop 2000 Spectrophotometer, a NanoDrop 3300 Fluorospectrometer, a BioRad CFX96 Touch Real-Time PCR Detection System, a Tecan Ultra plate reader, a Beckman Coulter refrigerated high-speed centrifuge, a Labconco CentriVap, a VWR UV transilluminator, an SI Analytics Lab 850 pH meter, two VWR analytical balances, a VWR Hotplate/Stirrer, one Thermo Scientific -80°C freezer, twelve -20°C freezers, a refrigerator, an E-C Apparatus Corporation EC-105 power supply, an ABN EPS-4000 power supply, a Pharmacia ECPS 3000/150 power supply, two small and two large (adjustable) vertical gel electrophoresis systems, three BioRad Mini-PROTEAN vertical gel electrophoresis systems, three horizontal gel electrophoresis systems, a Biometra temperature gradient gel electrophoresis (TGGE) system, a VWR heated water bath, VWR minicentrifuges, several sets of pipettes, vortexes, heating blocks, stir plates, a shaker, a microwave, a vacuum pump, computers, and all other supplies.
Each of the cell culture laboratories (BSL-1, BSL-2, and BSL-2 plus) is a 180 ft² space dedicated to basic cell culture techniques and cytotoxicity assays. Each laboratory space includes one sink and exceptional bench and drawer space. The room is equipped with two Labconco Logic Plus biosafety cabinets, two BVC professional fluid aspiration systems, two Thermo Scientific Heracell VIOS 160i CO2 incubators (37 °C, 5% CO2), a Leica DMIRE2 inverted fluorescent microscope, a Leica tissue culture microscope, a Tecan Spark microplate reader, a Attune NXT flow cytometer, a BD Accuri C6 Flow Cytometer, a Beckman Coulter Allegra X-30 high speed centrifuge, an Omni-Ruptor 4000 sonicator, a Life Technologies Countess II FL Automated Cell Counter, a Thermolyne Locator 8 Plus Cryo Biological Storage System, a refrigerator, VWR heated water bath, microcentrifuges, pipettes, vortexes, computers, and all other basic cell biology supplies.
Additionally, the equipment and room for work with bacteria include a Benchmark MyTemp Mini Digital Incubator, GeneMate Incubated Shaker, microcentrifuges, pipettes, vortexes, and all other basic cell biology supplies.
